Live-or-Dye™568/583isafixablereddye,designedfordiscriminationbetweenliveanddeadcellsduringflowcytometryormicroscopy.Live-or-Dye™568/583isacellmembraneimpermeableamine-reactivedye.Itisabletoenterintodeadcellsthathavecompromisedmembraneintegrityandcovalentlylabelfreeaminesonintracellularproteins.Thisdyelabelingisextremelystable,allowingthecellstobefixedandpermeabilizedwithoutlossoffluorescenceordyetransferbetweencells.
ThiazoleOrangeisagreenfluorescentdyethatlocalizestothecytoplasm,withsomenuclearaccumulation.Itisfixable,resultingindiffusecytoplasmicstaininginfixedcells.
Afterstainingyeastwiththiskit,bothlivinganddeadcellswillbestainedwithgreenfluorescence(Ex/Em512/533nm).Onlydeadcellswillbestainedwithredfluorescence(Ex/Em568/583nm),andmayappearyellowwhenthetwoimagesareoverlaid.
