
Ethidiummonoazidebromide(EMA)isafluorescentnucleicacidstainwithaphotoaffinitylabel.Thedye,afterphotolysis,bindscovalentlytonucleicacids.ThedyehasbeenusedtofootprintdrugbindingsitesonDNA2,tomodifyplasmidDNA,3,4andforlabelingyeastcellsforleukocytephagocytosisassays5.Aparticularlyusefulapplicationofthedyeistolabeldeadcellsinamixedpopulationofliveanddeadcells.BecauseEMA isrelativelyimpermeanttolivecells,itselectivelybindsDNAindeadcells.PhotolysisrendersthedeadcellDNAcovalentlylabeledwiththedye.Onecanthenwashandfixthecellpreparationforanalysisbymicroscopy,fluorescenceplatereaderorflowcytometry.Themajoradvantageofthismethodisthatresearcherscanavoidextensivemanipulationoflivepathogenicorganisms.7
EMA hasbeenusedtodifferentiatebetweenviableanddeadbacteriaby5′-nucleasePCR.8
