Live-or-Dye™FixableViabilityStainingKitsaredesignedfordiscriminationbetweenliveanddeadcellsduringflowcytometry.Live/deadstainsareusefulprobestoincludewhenanalyzingcellsurfaceproteinexpressionbyflowcytometry,becausetheyallowintracellularfluorescencesignalfromdeadcellswithpermeableplasmamembranestobeexcludedfromanalysis.
Live-or-Dye™FixableViabilityStainsarecellmembraneimpermeableamine-reactivedyes.Thedyesareabletoenterintodeadcellsthathavecompromisedmembraneintegrityandcovalentlylabelfreeaminesonintracellularproteins.Onlivecells,thedyesreactwithsurfaceproteins,butthesearemuchlessabundantthanintracellularproteins,resultinginlowstaininglevelscomparedtodeadcells.Thedyelabelingisextremelystable,allowingthecellstobefixedandpermeabilizedwithoutlossoffluorescenceordyetransferbetweencells.TheLive-or-Dye™stainingprotocolhasbeenoptimizedtomaximizelive/deaddiscriminationwithminimallivecellstaining,inordertopreventinterferencewithimmunostaining.
BiotiumoffersaselectionofeightdifferentLive-or-Dye™viabilitystainsspanningthefluorescencespectrum,formaximalflexibilityinmulti-coloranalysis.
