MitoView™dyesareavailableinthreecolors:MitoView™Blue,MitoView™Green,andMitoView™633.
MitoView™Blueisabluefluorescentmitochondrialdyewithabsorbance/emissionat398/440nm,suitablefordetectionbyconfocalmicroscopyorflowcytometryusingsettingsforDAPIorPacificBlue®.Thedyeismembranepermeableandbecomesbrightlyfluorescentuponaccumulationinthemitochondrialmembrane.Stainingisdependentonmitochondrialmembranepotential,andcanbeusedtomonitormitochondrialmembranepotentialinintactcells.Thedyeisdesignedforuseinlivecellsandisnotfixable.Note: MitoView™Bluemaynotbemembranepotential-dependentinyeastcells.
MitoView™GreenisagreenfluorescentmitochondrialdyewithpropertiessimilartothoseofMitoTracker®GreenFM.Thedyeisnonfluorescentuntilitpartitionsintothemitochondrialmembrane.Thestainingreliesonmitochondrialmass,notonmitochondriamembranepotential.Thus,thedyecanbeusedtostainmitochondriainbothlivecellsandfixedcells.MitoView™GreenisspectrallysimilartoFITC,makingitoptimallyexcitablebythe488nmargonlaserline.Note: MitoView™Greenappearstobesomewhatmembranepotential-dependentinyeastcells.
MitoView™633isafar-redfluorescentmitochondrialdyewithabsorbance/emissionat622/648nm.Thedyeismembranepermeableandbecomesbrightlyfluorescentuponaccumulationinthemitochondrialmembrane.Stainingisdependentonmitochondrialmembranepotential,andcanbeusedtomonitormitochondrialmembranepotentialinintactcells.Thedyeisdesignedforuseinlivecells,andisnotfixable. Note: MitoView™633maynotbemembranepotential-dependentinyeastcells.
MitoView633isalsoavailabletogetherwiththeNucViewcaspase-3substrateinadualapoptosisassaykit.
Note:TheoptimaldetectionsettingsforMitoView™633arethesameasforCy®5andotherfar-reddyes.However,thedyealsohasvisIBLeredfluorescenceandcanbeimagedusingCy®3settingsaswell.Asaconsequence,thedyecannotbeusedfortwo-colorimagingwithotherredprobes.
InadditiontoMitoView™dyes,Biotiumalsooffersseveralclassicaldyesformeasuringmitochondrialmembranepotential.TheseincludeJC-1,aratiometricdyethatformsredaggregatesinhealthycells,whicharereducedtothegreenmonomericstatewhenmitochondrialmembranepotentialislost.WealsoofferTMRMandTMRE,tworedfluorescentdyesthatareusedtoquantitativelymeasuremitochondrialmembranepotentialusingtheNernstequation.
